Blood Agar Base with Low pH (500gm) by HiMedia is a high-quality microbiological culture medium designed for the isolation and cultivation of fastidious pathogenic microorganisms. Laboratories commonly use this medium as a basal agar for preparing blood agar plates by adding sterile defibrinated blood.

This specialized formulation has a low pH of approximately 6.8, which helps stabilize red blood cells and improves the visualization of hemolytic reactions. As a result, microbiologists can easily observe clear hemolysis zones produced by organisms such as Streptococcus and Neisseria. Therefore, this medium plays an important role in clinical microbiology and diagnostic laboratories.

Additionally, laboratories can use the medium without adding blood as a general-purpose culture medium. Because of its balanced nutrient composition, it supports the growth of a wide range of microorganisms.

      Key Features

      • Optimized Low pH (6.8) – Stabilizes red blood corpuscles and enhances hemolysis detection

      • Ideal Basal Medium – Used for preparing blood agar by adding sterile blood

      • Supports Fastidious Microorganisms – Suitable for organisms like Neisseria and Streptococci

      • Clear Hemolysis Observation – Helps identify alpha, beta, and gamma hemolysis

      • High-Quality Microbiological Media – Manufactured by HiMedia

      • Versatile Laboratory Use – Can also function as a general-purpose medium

      Applications

Blood Agar Base with Low pH is widely used in clinical microbiology, diagnostic laboratories, and research facilities.

      Common applications include:

      • Isolation of fastidious pathogens from clinical samples

      • Cultivation of Streptococci and Pneumococci

      • Observation of hemolytic reactions on blood agar plates

      • Detection of phosphate-producing staphylococci when used with phenolphthalein phosphate

      • Determination of the salinity range of marine Flavobacteria

      • Preparation of Salmonella Typhi antigens

Furthermore, the medium provides excellent nutrient support for many bacterial species when used with or without blood supplementation.

      Preparation Method (General)

      1. Dissolve the required amount of dehydrated medium in distilled water.

      2. Heat with gentle stirring until the powder completely dissolves.

      3. Sterilize by autoclaving at 121°C for 15 minutes.

      4. Cool the medium to 45–50°C and aseptically add sterile defibrinated blood if preparing blood agar plates.

      5. Mix gently and pour into sterile Petri dishes.

      Storage Instructions

      • Store the container tightly closed in a cool and dry environment.

      • Protect from moisture and direct sunlight.

      • Use prepared media promptly for best microbiological results.
